Product Overview:

Mammalian Thioredoxin reductase (TrxR) is is a FAD-containing enzyme of the Thioredoxin system that catalyzes the conversion of oxidized Trx to reduced Trx using reducing equivalents from NADPH. Whereas bacterial TrxR is specific for reduction of bacterial Trx, mammalian TrxR reduces many other substrates in addition to Trx from different origin. Thioredoxin maintains the redox pool of the cell, and is known to be induced in conditions of oxidative stress. Thioredoxin is also an anti-oxidative protein that has been shown to provide protection against oxidative stress in cardiovascular diseases, neurodegenerative conditions, and other inflammatory diseases. Increased expression of thioredoxin is known to be associated with increased proliferation of cancer cells. Therefore, TrxR is a very important protein that maintains the redox environment in reduced state by reducing Thioredoxin.

Application:

In our assay method 5,5-dithiobis (2-nitrobenzoic acid) is used as a substrate for reduction by mammalian TrxR. The product of the reaction, TNB (5’-thionitrobenzoic acid) is yellow and has an absorbance maximum at 412 nm with an extinction coefficient of 13,600 M-1 cm-1. The assay measures the formation of TNB at an absorbance maximum of 412 nm, one unit of TrxR activity is defined as increase in absorbance of 1 at A412 nm. In the assay, the activity of TrxR is inhibited by a specific inhibitor of TrxR. Therefore, specific reduction of DTNB by TrxR is calculated by deducting inhibition-dependent reaction rate from total reaction rate that provides specific rate for TrxR.
